DELFIA TRF Assays
DELFIA® (dissociation-enhanced lanthanide fluorescent immunoassay) TRF technology combines the benefits of time resolved fluorescence (TRF) with the use of lanthanide chelates for a highly sensitive, robust and reproducible assay format. With over 20 years of history, and thousands of publications, DELFIA is ideal for simple to complex matrices and can be used for both biochemical and cell-based assays.
The use of long-lifetime fluorescent lanthanide chelates, rather than other fluorophores, allows for a delay in measurement between excitation and emissions, which occur simultaneously in standard fluorometric assays. By combining the benefits of TRF with the use of lanthanide chelates, DELFIA offers multiple advantages over standard fluorometric detection:

This kit has the same assay principle as a chromium release assay, with target cells loaded with a BATDA ligand instead of 51Cr. There is no requirement for specially-engineered cells.

Europium-labeled anti-HA antibody for binding HA-tagged proteins in DELFIA® time-resolved fluorescence (TRF) assays. The anti-HA antibody is a purified IgG2b subclass of mouse monoclonal antibody. It recognizes the epitope sequence (YPYDVPDYA) derived from the human influenza hemagglutinin (HA) protein.

The DTA-activated chelate reacts with free amino groups, sulfhydryl groups, and hydroxyl groups. Because of its high reactivity, we usually do not recommend DTA as it may “inactivate” the reagent you are labeling.
