Omni Bead Ruptor Bead Mill Homogenizer Protocols for Tissue Homogenization and Cell Lysis

Various tissues and sample types were homogenized and assessed for analytes using the portfolio of Omni Bead Ruptor bead mill homogenizers. Below is our list of resulting performance-verified protocol snapshots, for tissue and cell homogenization protocols and specific bead mill homogenizer protocols. Note that all listed protocols are only recommended starting points from our or external lab experiences, and labs will need to verify their own protocols for their own workflows.

When comparing several commercially available bead mill homogenizers based on maceration level, nucleic acid quality, quantity, amplification, and DNA shearing to determine which instruments were effective for their sample type, the authors’ wrote:

“Upon receiving the highest score on the evaluation matrix, the Bead Ruptor 24 Elite was further tested to determine whether it could consistently disrupt a tick body enough to release detectable quantity of pathogen…. In the present study, the tested traditional bead homogenizer, the Bead Ruptor 24 [Elite], seemed to perform the best overall.”1

Several tissue disruption methods exist to support labs access the molecular analyte they need for their downstream assays and analyses. Some rely on chemical lysis of tough materials and membrane disruption while others rely on mechanical disruption using shearing or impact force. Homogenizers fall into the latter category – leveraging rotor-stator or bead-milling forces to uniformly homogenize starting material. As mechanical disruption does not require the harsh use of chemicals, some researchers prefer to rely on homogenization for either all or part of their tissue disruption methods. These researchers require thorough cell lysis, but wish to keep cellular stress to a minimum, as well as avoid excessive handling of toxic chemicals for their lab personnel.
