Assay validation

Evaluation of the two PPI Eu buffers in protein/protein interaction assays

The two Europium PPI detection buffers were evaluated in two different Protein/Protein interaction assays.

The IC50 values corresponding to the inhibition of the two interactions (A/B & C/D) remained comparable in both PPI Eu detection buffers.

The Protein A/ Protein B interaction displayed the same Signal to Background (S/B) in the PPI Eu detection buffer and the PPI Low Ionic Strength Eu detection buffer.

However, the S/B of the Protein C/ Protein D interaction was significantly improved in the new PPI Low Ionic Strength Eu detection buffer, suggesting that the PPI Low Ionic Strength Eu detection buffer can maximize assay performance when the studied protein-protein interaction is sensitive to high ionic strength.
