Overview

AlphaPlex-545 (Terbium) Acceptor beads conjugated to antibody against human Histone H3 that is unmodified (H3K4), for use in Alpha no-wash multiplexing epigenetic assays.

Alpha technology is a no-wash homogeneous proximity immunoassay that includes Alpha Donor and AlphaPlex-545 Acceptor beads to quantitate activity on histone, peptide, or protein substrates. In vitro modification of substrate is performed by incubating enzyme with appropriate cofactor and histone substrate. Following the enzymatic reaction, methylation or acetylation is monitored using specific antibody-conjugated AlphaPlex Acceptor beads. Following enzymatic reaction,streptavidin-coated Donor beads are added to capture the substrate, the beads are brought into close proximity in the absence of epigenetic mark on Histone H3 (Lysine 4) on the target substrate. The Donor beads are excited by a laser at 680 nm to release singlet oxygen. The singlet oxygen causes a reaction in the Acceptor bead and light emission is produced at 545 nm. This emission is proportional to the amount of unmodified / non-methylated / non-acetylated substrate present.

AlphaPlex-545 Acceptor beads are intended for use in multiplexing Alpha assays, in conjunction with Alpha Donor beads and AlphaLISA™ Acceptor beads.

  • 250 µg of Acceptor beads is sufficient to run ~500 wells using a 50 µL reaction volume.
  • 5 mg of Acceptor beads is sufficient to run ~10,000 wells using a 50 µL reaction volume.
  • 25 mg of Acceptor beads is sufficient to run ~25,000-50,000 wells using a 50 µL reaction volume.
