AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
Stem cell factor (SCF), also known as c-kit ligand, is a 28‑40 kDa transmembrane glycoprotein expressed in many cell types. SCF promotes the survival, differentiation, and mobilization of multiple cells. Mature human SCF consists of a 189 amino acid extracellular domain, a 23 amino acid transmembrane segment, and a 36 amino acid cytoplasmic tail. Non-covalent dimers of transmembrane or soluble SCF interact with the receptor tyrosine kinase SCF R/c‑kit to trigger receptor dimerization and signaling to become a primary growth and activation factor for mast cells and eosinophils. Clinically, SCF assists in the recovery of cardiac function following myocardial infarction by increasing the number of cardiomyocytes and vascular channels. Additionally, there are more than 75 on going clinical trails involving SCF.
