The AlphaLISA® immunoassay kit for human phosphorylated neurofilament heavy chain protein (pNFH) enables the quantitative determination of human pNFH in plasma or cerebrospinal fluid (CSF) using a homogeneous AlphaLISA assay (no wash steps).

AlphaLISA technology allows the detection of molecules of interest in buffer, cell culture media, serum and plasma in a highly sensitive, quantitative, reproducible and user-friendly mode. In an AlphaLISA assay, a Biotinylated Anti-Analyte Antibody binds to the Streptavidin-coated Alpha Donor beads, while another Anti-Analyte Antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads provokes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
Phosphorylated neurofilament heavy chain protein (pNFH) is also known as NF-200; NF200; NEFH; N52; Neurofilament heavy polypeptide; Neurofilament triplet H protein; 200 kDa neurofilament protein; KIAA0845. It is a 200 kDa subunit of the three so-called neurofilament triplet proteins: light (68 kDa), medium (160 kDa) and heavy (200 kDa), which are major structural components of neurons. High levels of neurofilament have been observed during the progression of numerous neurological diseases. Many studies have demonstrated that pNFH can be used as a biomarker to monitor neurodegenerative diseases such as Amyotrophic Lateral Sclerosis (ALS) and ischemic stroke.
