Interleukin-18 (IL-18) is an 18 kDa cytokine produced by Kupffer cells, activated macrophages, keratinocytes, intestinal epithelial cells, osteoblasts, and adrenal cortex cells. IL-18 is involved in the production of interferon-γ (IFN-γ) in response to toxic shock and shares functional similarities with IL-12. IL-18 works together with IL-12 to induce cell-mediated immunity following infection with microbial products like lipopolysaccharides (LPS). IL-18 is also able to induce severe inflammatory reactions, which suggests a role in certain inflammatory disorders. Elevated plasma levels of IL-18 have been reported in hematological malignancies, inflammatory diseases, and autoimmune disorders.
AlphaLISA™ technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
