Alpha-1 Antitrypsin (A1AT) is a 52 KD protein. The serpin superfamily includes the protease inhibitors alpha-1 antitrypsin and antithrombin, which are widely studied due to their involvement in the blood coagulation and inflammation. A deficiency in antitrypsin causes emphysema ((Chronic obstructive pulmonary disease (COPD), also known as chronic obstructive lung disease (COLD), and chronic obstructive airway disease (COAD)). Circulating levels of A1AT is an excellent indicator of patients with inflammatory diseases. The present kit detects hA1AT in human serum, plasma, and cell culture supernatants.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
