AlphaLISA technology allows the detection of molecules of interest in buffer, cell culture media, serum and plasma in a highly sensitive, quantitative, reproducible and user-friendly mode. In an AlphaLISA assay, a Biotinylated Anti-Analyte Antibody binds to the Streptavidin-coated Alpha Donor beads, while another Anti-Analyte Antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads provokes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
Galectin-9 belongs to a large family of beta-galactoside-binding proteins. Galectin-9, also known as LGLAS9, Ecalectin and UAT, is a mammalian lectin with a molecular weight around 50 kD. Galectin-9 is expressed in a variety of cells such as T cells, PMNs, astrocytes, fibroblasts and endothelial cells. Galectin-9, as an immuno oncology checkpoint biomarker, induces cellular apoptosis in part by direct binding to TIM-3. It is also involved in cell aggregation, adhesion, and chemotaxis; it also induces regulatory T cells and suppresses Th1 and Th17 responses. This kit has been designed for the detection of human Galectin-9 in serum, cell lysate, and cell culture supernatants.
