AlphaLISA technology allows the detection of molecules of interest in buffer, cell culture media, serum and plasma in a highly sensitive, quantitative, reproducible and user-friendly mode. In an AlphaLISA assay, a Biotinylated Anti-Analyte Antibody binds to the Streptavidin-coated Alpha Donor beads, while another Anti-Analyte Antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads provokes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
Cluster of differentiation 80 (CD80), also known as B7-1, is a protein that is activated by the binding of CD28 or CTLA-4, providing a costimulatory signal necessary for T cell activation and survival. This protein is expressed on dendritic cells, macrophages, and activated B cells. It has been shown that CD80 and CD86 can substitute for one another in the initial activation of resting CD4(+) T cells as well as the maintenance of their proliferative response. Studies show that increased levels of CD80 in urinary excretion have been linked to Fabry disease, and Minimal Change Disease. This kit is designed to detect and quantify the levels of CD80 in cell culture supernatant and serum.
