VAV1 is a Rho-family guanine nucleotide exchange factor that is encoded by a proto-oncogene, Vav Guanine Nucleotide Exchange factor 1. Wild-type VAV1 is recognized as a cardinal signal transducer protein in the hematopoietic system, where it is exclusively expressed. However, mutations in various domains of the VAV1 protein are present in human cancers, including T-cell leukemia/lymphoma, peripheral T-cell lymphomas, lung adenocarcinoma, and squamous cell carcinomas. VAV1 is also an activator of T and B cell receptor signaling, therefore making it a potential target for protein degradation therapies to combat autoimmune disease, such as multiple sclerosis.
