Overview

Ubiquitin is a highly conserved regulatory protein found in most tissues of eukaryotic organism. Phosphorylation of ubiquitin is a key step in the process of mitochondria quality control through the Parkin/PINK-1 interaction. Parkin is a ubiquitin ligase E3 that conjugates ubiquitin to impaired-mitochondrial proteins, leading to organelle degradation. PINK1 is a Ser/Thr kinase that accumulates only on impaired mitochondria and phosphorylates two substrates, the ubiquitin-like domain of Parkin and ubiquitin chains. The amplified Parkin/PINK-1 dependent ubiquitin functions act as a signal for mitophagy. Dysregulation of this signaling is linked to neurodegenerative diseases such as Parkinson's Disease.

Formats:
  • The HV (high volume) kit contains reagents to run 100 wells in 96-well format, using a 60 μL reaction volume.
  • The 500-point kit contains enough reagents to run 500 wells in 384-well format, using a 20 μL reaction volume.
  • The 10,000-point kit contains enough reagents to run 10,000 wells in 384-well format, using a 20 μL reaction volume.
  • The 50,000-point kit contains enough reagents to run 50,000 wells in 384-well format, using a 20 μL reaction volume.
