Transcription factor EB (TFEB) plays a key role in the regulation of basic cellular processes, including lysosomal biogenesis and autophagy. While phosphorylated TFEB is found in the cytoplasm, dephosphorylated TFEB moves to the nucleus to initiate the transcription of target genes. The subcellular location and function of TFEB are regulated by phosphorylation at the lysosomal surface, which is controlled by mechanistic target of rapamycin (mTOR). TFEB is implicated in various diseases, such as lysosomal storage disorders, neurodegenerative diseases, cancers, and metabolic disorders.
