SMAD family member 4 (SMAD4), also known as mothers against decapentaplegic homolog 4, or DPC4 (deleted in pancreatic cancer-4), belongs to the SMAD family of transcription factor proteins, which act as mediators of TGF-β signal transduction. In conjunction with SMAD 2 and SMAD 3, the SMAD2/3/4 complex forms and acts as a transcription factor that promotes the expression of a pro-fibrotic phenotype. Mutated SMAD4 is found in many types of cancers and fibrosis.
