PTEN-induced kinase 1 (PINK1) is a mitochondrial serine/threonine-protein kinase, closely involved in mitochondrial quality control by identifying damaged mitochondria and targeting specific mitochondria for degradation. In damaged depolarized mitochondria, PINK1 accumulates on the mitochondrial outer membrane. Upon activation, PINK1 activates Parkin which will ubiquitinate many targets at the Mitochondrial Outer Membrane (MOM). One of these targets is TOM20. Dysregulated PINK1 is associated with Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, ALS, liver disorders, and kidney disorders.
