Parkin, encoded by the PARK2 gene, is an E3 ubiquitin ligase which has a neuroprotective role in combination with the Ser/Thr kinase PINK1 by acting in a mitochondrial quality control pathway. Since mitochondria are continuously exposed to intra and extra mitochondrial threats including reactive oxygen species (ROS), the removal of damaged mitochondria is critical for maintaining cellular homeostasis. Dysfunction of Parkin represents both a predominant cause of familial parkinsonism but also a formal risk factor for the more common, sporadic form of Parkinson's disease (PD).
