General control nonderepressible 2 (GCN2) is a serine/threonine-protein kinase that plays a key role in modulating amino acid metabolism as a response to nutrient deprivation. GCN2 is one of the four known sensors of the integrated stress response (ISR), which is a cytoprotective response mediated by eIF2α. Under conditions of amino acid deprivation, GCN2 inactivates eIF2α by phosphorylation, and eIF2α phosphorylation can also lead to enhanced transcription of mRNA molecules involved in host response to infection and inflammation. GCN2 is associated with cancer.
