Fibroblast growth factor receptor 3 (FGFR3) is a transmembrane Receptor Tyrosine Kinase consisting of one intracellular tyrosine kinase domain. Upon ligand binding, the receptor dimerizes, and transphosphorylation of tyrosine kinase domains occurs. Activation of downstream signaling occurs via FRS2, which functions as a key adaptor protein, resulting in MAPK and PI3K/AKT activation. Genetic alteration of FGFR3 leads to constitutive activation of this receptor and has significant consequences in carcinogenesis, involved in lymphomas, urothelial, hepatocellular, lung, breast, and colorectal cancers.
