Cyclin-dependent kinase 12 (CDK12) is a transcription-associated CDK. It complexes with Cyclin K to regulate gene transcription elongation via phosphorylating RNA polymerase II and regulates the expression of genes involved in DNA repair. CDK12 also plays a role in RNA splicing, cell cycle progression, cell proliferation, DNA damage response (DDR), and the maintenance of genomic stability. Mutation or amplification of CDK12 is closely related to tumorigenesis, associated with cancers such as breast, colorectal, liver, lung, ovarian, prostate, and bladder.
