Beclin-1 (BECN1) plays a central role in the regulation of autophagy and cell death. Together with PI3KC3, Beclin-1 nucleates autophagosome formation and is essential for proper degradation of cellular components. The activity of Beclin-1 is impacted by many factors, especially different binding partners. Numerous membranic, cytosolic, and nuclear proteins have been identified as activators of autophagy by binding to Beclin-1. Improper Beclin-1 signaling is associated with cancer, neurodegenerative diseases, and cardiac dysfunction.
