Overview

IVISense 680 NHS fluorescent labeling kit is designed for preparing fluorescently labeled antibodies, proteins, or peptides for in vivo imaging applications in small animals. Each kit contains our superior in vivo optimized IVISense 680 NHS Fluorescent Dye fluorophore and everything else you need for carrying out the labeling reaction and purifying the labeled product. The reactive fluorophore has a succinimidyl ester group, which reacts with an amine group on the protein (i.e., lysine side-chain amine) to form a stable amide linkage. IVISense 680 NHS Fluorescent Dye is supplied in two separate vials, each containing adequate material for labeling 0.5-5 mg of an antibody. Following the labeling reaction, the unconjugated fluorophore is conveniently and rapidly removed by purification columns with a molecular weight cutoff of 7 kDa.

  • 2 X 0.25 mg of IVISense 680 NHS Fluorescent Dye
  • 1 X 1 mL of 1M solution of sodium bicarbonate (pH8.3).
  • 2 X Purification column
  • 4 X 15 mL conical collection tube
  • 1 X PBS (50 mL)

IVISense™ 680 fluorescent dyes are ideal for molecular labeling for use in preclinical in vivo imaging applications and in vitro corroboration assessments by microscopy and flow cytometry. IVISense 680 dyes are well suited for the labeling and imaging of proteins, antibodies, and small molecules in a wide range of research areas. The 680 nm fluorescence wavelength is ideal for biological imaging applications using in vivo fluorescence systems and provides sufficient signal in most advanced microscopes, high content imaging and flow cytometry.

  • Uniquely optimized for the generation of in vivo imaging probes with low non-specific accumulation at tumor sites and inflammatory sites.
  • Suboptimal but sufficient excitation and emission with light sources and filters in all commonly used microscopes, flow cytometers, and plate readers.
  • Suitable for in vivo multiplexing with 750 or 800 nm Revvity probes or other labeled proteins/Abs. Expanded in vivo multiplexing with IVIS spectral unmixing with 645/750/800 nm probes or antibodies.
  • Requires maintaining mice on alfalfa-free diet to remove spectral overlap with diet autofluorescence.
  • Recommended dye:protein ratio of 1-3.

IVISense 680 NHS 673/691 When a protein is labeled with multiple IVISense Self-Quenching Dye molecules, there is a shift in the absorbance of the dye, which will reduce the signal.

Only choose IVISense Self-Quenching Dyes if your labeling ratio of dye:protein will be 1:1. If multiple dyes are to be conjugated onto your protein, a non-self-quenching IVISense Dye should be used. 

NHS and MAL dyes are suitable as noted above.
