IVISbrite Bioluminescent Substrates

Overview

Revvity offers two IVISbrite™ bioluminescent luciferase substrates for in vitro, in vivo and ex vivo bioluminescent imaging: D-luciferin and coelenterazine H. D-luciferin is a substrate for Firefly luciferase (such as Red-FLuc), and is used in conjunction with our IVISbrite Tumor Cell Lines or cells transfected with our IVISbrite Red F-luc-Puromycin (formerly RediFect™ Red-FLuc) and IVISbrite Red F-luc-GFP (formerly Red-FLuc-GFP) lentiviral particles for in vitro, in vivo or ex vivo imaging. Coelenterazine H is a substrate for Renilla and Gaussia luciferase and can be used for in vivo imaging with cells transfected with our IVISbrite Green Renilla-Puromycin (formerly RediFect Green RenLuc-Puromycin) lentiviral particles, or when either of these luciferases is used as a reporter. The respective substrate is necessary for bioluminescent visualization in vitro, in vivo or ex vivo.

After the bioluminescent cell line expressing firefly luciferase, Renilla luciferase, or Gaussia luciferase is established in a mouse or other small animal model, the luciferin or coelenterazine substrate is injected. The substrate is then used by the bioluminescent reporter to produce light within the cell line, which allows visualization of the bioluminescent cells within the mouse (or other) model. This luminescence can be visualized and measured using an appropriate 2D or 3D imager.

FAQs

Q. How do I know whether to use the intraperitoneal (i.p.) luciferin injection protocol or the intravenous (i.v.) injection protocol, when injecting luciferin for bioluminescence in vivo imaging?

 A. Most scientists use i.p injections. Subcutaneous injections are sometimes performed as well. If the signal is weak, you can do an i.v. injection, but you will need to image as soon as you inject (which can be very cumbersome). 

Q. What if I was using product number 122796, 119222, or XR-1001 previously? 

A. These products have been replaced by product number 122799. They are all firefly D-luciferin, potassium salt formulations.
